Description	Antibody/Agonist/ADC Effect Reporter Lentivirus. Series of lentiviral particles for evaluating various cellular processes such as ADCC, ADCP, STING pathway activity, anti-PD1/PDL1 antibodies, don t eat me signaling and more. Make your own cell line and monitor transcriptional activity in response to different stimuli. Free yourself from the constraints of fixed commercial cell lines and make your own. Experience the flexibility to choose from a variety of fluoresecent and luminescent dual reporters and drug selection markers. All of the products feature an all-in-one lentiviral expression construct that combines a constitutively expressed surface receptor or protein (e.g. PD-1, CD16, CD32, etc.) and a fluorescent + secreted GLuc dual reporter that is conditionally expressed in response to transcriptional activity of a particular transcription factor (e.g. NFAT, NFkB, etc.). The cDNA encoding the receptor or protein is constitutively expressed by this construct under control of EF1a promoter. Downstream of PD-1 is a reporter gene or selection marker that is also expressed by EF1a, but separated from receptor/protein gene by P2A (separate proteins are expressed from a single mRNA transcript). Separately, a minimal TATA promoter coupled with tandem repeats of the particular transcriptional response elements drive expression of a second reporter to read out the TF transcriptional activity, which can be used as an indicator of T cell activation, ADCC, ADCP, etc. A proprietary enhancer element is situated upstream of the response elements to boost the signal intensity. All components are contained within a single construct pre-packaged in high-titer lentiviral particles, allowing you to generate your own stable cell lines with ease. The dual reporter format allows for the convenient measurement of cellular processes such as ADCC by bioluminescence or fluorescence. The reporter lentivirus is purified by PEG precipitation and sucrose gradient centrifugation, and is ideal for use in difficult-to-transfect cells including primary and/or thawed cells.
