Tag
N-terminal FLAG-tag, C terminal His tag
Formulation
40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, 0.04% Tween-20, 20% glycerol, and 3 mM DTT.
Species
Human
Application
Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling
Notes
Add 50 ul reaction mix containing a-ketoglutarate, TET enzyme, ATP, and TET SA assay buffer (BPS) to wells coated with the substrate. TET SA buffer contains saturable concentrations of iron, a-KG, and ATP. Incubate 3hrs at 37°C. Add antibodyagainst hydroxymethylated cytosine, incubate 1 hr. Then add secondary HRP labeled antibody and incubate 30 min. Finally, add HRP chemiluminescent substrates and read luminescence.
