Reaction wasperformed with 20 mM HEPES (pH 7.4),150 mM NaCl, 500 uM -ketoglutarate, 25uM iron, 2 mM ascorbic acid, 0.01%Tween-20, 0.5 uM biotinylated peptidesubstrate, and JHDM1D added to thewells. Add antibody against methylatedH3K9. Incubate for 30 min followed byAlphascreen detection.
Avoidfreeze/thaw cycles. Storing dilutedenzyme is not recommended, ifnecessary, use carrier protein (BSA 0.1- 0.5%).
