Description	Thrombin (EC3.4.21.5, β-thrombin) is the product of proteolyticactivation of the zymogen prothrombin. Human thrombin is atwo-chain serine protease with a mass of 37 kDa. The active siteis located within the heavy chain. Thrombin has a high specificityfor certain arginine bonds in protein substrates. The primarysubstrate is fibrinogen which thrombin converts to fibrin throughthe cleavage of four arginyl-glycyl peptide bonds. Thrombin isalso an important activator of platelets, factor XIII, Protein C andTAFI (Plasma procarboxypeptidase B). In a positive feedbackmechanism, thrombin increases the rate of its own production byactivation of factors VIII and V. The rate of thrombin production issubsequently limited indirectly through the activation of Protein Cby thrombin, which then inactivates the activated cofactors VIIIand V. The binding of thrombin to thrombomodulin on the cellsurface dramatically alters thrombin™s specificity, increasing it™sactivity toward Protein C and TAFI, and decreasing it™s activitytoward fibrinogen and activating cofactors VIII and V. In plasma,thrombin activity is inhibited primarily by antithrombin and to alesser extent heparin cofactor II. The rate of inhibition by both ofthese inhibitors is profoundly increased in the presence ofoptimal concentrations of heparin. Other physiological inhibitorsof thrombin in the absence of heparin include β2-macroglobulinand β1-antitrypsin1-4.
