Single-Stranded DNA Binding Protein, SSB (5 ug/uL)

The Single-Stranded DNA Binding Protein (SSB) is a thermostable SSB protein specifically responsible for binding to single-stranded regions of DNA. It primarily binds to the single-stranded areas produced by helicase as it moves forward along the replication fork, preventing the newly formed single-stranded DNA from re-annealing to form double-stranded DNA or being degraded by nucleases. After binding to single-stranded DNA, SSB makes the helical dimers unstable, thus allowing DNA polymerase to more easily access the substrate.The PCR reaction can be optimized by adding SSB single-stranded binding protein to the reaction mixture. SSB can be added during the PCR process to keep the denatured DNA stable, or it can be used as a PCR reaction additive to protect ssDNA from being digested by nucleases. Studies have shown that SSB can improve PCR performance on complex templates by directly or indirectly acting on the polymerase, increasing strand displacement activity, and affinity for primer templates.
