Vaccinia Capping Enzyme ELISA Kit

In eukaryotes, mRNA undergoes post-transcriptional modifications, forming a special structure at the 5' end known as the cap structure. This structure plays an important role in mRNA stability, transport, and translation. The vaccinia virus capping enzyme is an effective enzyme that catalyzes the formation of the cap structure. It consists of two subunits, D1 and D12, and has RNA triphosphatase activity, guanylyltransferase activity, and guanine N7-methyltransferase activity. It can attach the 7-methylguanosine cap structure (m7Gppp) to the 5' end of RNA (m7Gppp5'N). 

The vaccinia virus capping enzyme, in the presence of capping buffer (Capping buffer), guanosine triphosphate (GTP), S-adenosylmethionine (SAM), and other components at appropriate concentrations, is capable of capping RNA within an hour while ensuring the correct directionality.

This kit uses the principle of a double antibody sandwich enzyme-linked immunosorbent assay (ELISA) to detect the residual amount of vaccinia virus capping enzyme. The vaccinia virus capping enzyme standard and test samples are added to a pre-coated enzyme plate with anti-vaccinia virus capping enzyme antibodies (36709-A), followed by the addition of a diluted biotin-labeled capping enzyme detection antibody (36709-C), and finally Streptavidin-HRP (SA-HRP) (36709-D). This forms the antibody+antigen+antibody-Biotin+SA-HRP complex. After washing, TMB substrate solution (36709-H) is added to develop color.

Under the catalysis of HRP enzyme, TMB is transformed from colorless to blue, and under the action of the stop solution (36709-I), is finally converted into yellow. The intensity of the yellow color is positively correlated with the amount of vaccinia virus capping enzyme detected in the sample.
