Inorganic Pyrophosphatase (PPase) catalyzes the conversion of one molecule of pyrophosphate into two molecules of phosphate ions. In nucleic acid amplification experiments, PPase can hydrolyze the inorganic pyrophosphate generated during the reaction. This is a highly exergonic reaction that can shift the reaction equilibrium towards the product side, thereby preventing inhibition of the reaction system.

In the production process of mRNA vaccine products, adding PPase can improve the yield of mRNA. However, for products with mRNA as the final product, the added PPase is an exogenous substance that needs to be removed during the purification process. Therefore, it is necessary to detect the residual amount of PPase to evaluate the effectiveness of the purification process in removing protein residues and to confirm whether the residual PPase in the mRNA vaccine product has been purified to an acceptable level.

This kit uses the principle of a double antibody sandwich enzyme-linked immunosorbent assay (ELISA) to detect PPase residues. PPase standard and test samples are added to a pre-coated enzyme plate with anti-PPase antibodies (36706-A), followed by the addition of a diluted biotin-labeled detection antibody (36706-C), and finally Streptavidin-HRP (SA-HRP) (36706-D). This forms the antibody+antigen+antibody-Biotin+SA-HRP complex. After washing, TMB substrate solution (36706-H) is added to develop color.

Under the catalysis of HRP enzyme, TMB is transformed from colorless to blue and, under the action of the stop solution (36706-1), is finally converted into yellow. The intensity of the yellow color is positively correlated with the amount of PPase detected in the sample.
