The principle is that starch hydrolase catalyzes the hydrolysis of starch to produce reducing sugar, which reduces 3, 5-dinitrosalicylic acid to produce brown-red substance with an absorption peak at 540 nm. Amylase activity was calculated by measuring the absorbance increase rate at 540 nm.
