The breast cancer susceptibility gene, BRCA1, which maps to chromosome 17q is expressed in numerous tissues, including breast and ovary. A second breast cancer susceptibility gene, BRCA2, located on chromosome 13q12-13, also confers a high incidence of breast and ovarian cancer. Mutations within the BRCA1 gene may account for at least 80% of families with increased incidence of both early-onset breast cancer and ovarian cancer. BRCA-AP1, also known as BAP1 (BRCA1 associated protein 1), localizes to the nucleus and interacts with the RING finger domain of the breast cancer 1, early onset protein (BRCA1). This gene is thought to be a tumor suppressor gene that functions in the BRCA1 growth control pathway.
